Hitman 3 features Contracts Mode, where a player can select up to five targets across all the maps in the series, add rules and complications like specific kill methods and share their contracts with other players. Unlike Hitman 2, there are no multiplayer modes, with Sniper Assassin being solely single-player, and Ghost Mode being removed entirely. The VR functionality retroactively applies to levels imported from the previous two entries.

The game features PlayStation VR compatibility on the PlayStation 4 version, with support for PlayStation 5 via backwards compatibility, Support for VR on PC was added on 20 January 2022. Another new feature in the game is numerical keypads, where players have to find four-digit codes and enter them manually to open doors and safes. Hitman 3 added a new camera item available by default which can be used to take photos and is used for gameplay purposes such as opening doors on certain maps. Each stage includes several initially locked doors which can only be unlocked from one side once unlocked, these doors stay open in all future playthroughs, allowing initially closed-off locations to be reached more quickly. The new locations in Hitman 3 include persistent shortcuts, a new gameplay feature to the series. Owners of Hitman or Hitman 2 are able to import maps, levels, and their progress into Hitman 3.

A seventh map, the fictional Ambrose Island in the Andaman Sea, India, was added for free on 26 July 2022. The base game features six new locations: Dubai, Dartmoor, Berlin, Chongqing, Mendoza, and the Carpathian Mountains, Romania. In the game, 47 travels to various locations and carries out contracted assassinations, continuing the story of the last two games. Like its predecessors, Hitman 3 is a stealth game played from a third-person perspective and players once again assume control of assassin Agent 47. This rebranding included the previous two Hitman games becoming available to Hitman 3 owners, free of charge, along with a new game mode called "Freelancer". In 2023, IO rebranded Hitman 3 as Hitman: World of Assassination, which was previously used in the Stadia version. Hitman 3 was also the most commercially successful game in the series, and was extensively supported by IO with several releases of downloadable content and free updates that added new features, game modes, and a new location. The game won various awards including "PC Game of the Year" at the 2021 Golden Joystick Awards.

Some critics called it the best entry in the series and it has been cited as one of the greatest stealth games of all time. It received positive reviews, with praise for its level design and atmosphere, stealth mechanics, and 47's abilities. Hitman 3, the first game to be self-published by IO Interactive after becoming an independent studio, was released worldwide for PlayStation 4, PlayStation 5, Windows, Xbox One, Xbox Series X/S, Stadia, and Nintendo Switch (via cloud gaming) on 20 January 2021.
Each mission also presents challenges that players can complete to unlock new items.
Like its two predecessors, the game is structured around six levels, five of which are large sandbox locations that players can freely explore to find opportunities to eliminate their targets. The game is presented from a third-person perspective, with a focus on interactive elements in 47's environment. Concluding the story arc started in 2016's Hitman, the game follows genetically-engineered assassin Agent 47 and his allies as they hunt down the leaders of the secretive organization Providence, which controls global affairs and was partially responsible for 47's creation and upbringing. It is the eighth main installment in the Hitman video game series, the sequel to 2018's Hitman 2, and the third and final entry in the World of Assassination trilogy. Hitman 3 is a 2021 stealth game developed and published by IO Interactive.
